GLP-1 and Dual Agonists: Mechanism Spotlight

GLP-1 (glucagon-like peptide-1) is an incretin hormone secreted by intestinal L-cells in response to food intake. It enhances insulin secretion, slows gastric emptying, and increases satiety.

GLP-1 receptor agonists such as Semaglutide bind the GLP-1 receptor with high affinity and resist enzymatic degradation, giving them extended in vivo activity.

Tirzepatide adds a second arm: activation of the GIP (glucose-dependent insulinotropic polypeptide) receptor, producing dual incretin signaling.

Research into these dual agonists is expanding rapidly, with endpoints spanning glycemic control, weight regulation, and cardiometabolic markers.
